喘可治注射液对COPD患者外周血单个核细胞分泌TNF-α的影响

摘　　要：目的探讨喘可治注射液(下称喘可治)对慢性阻塞性肺疾病(COPD)稳定期患者外周血单个核细胞(PBMCs)分泌肿瘤坏死因子-α(TNF-α)的影响。方法稳定期COPD患者和健康对照者各20例,均用密度梯度法分离患者和健康对照者的PBMCs,加入脂多糖(LPS)和4%喘可治,并与空白培养、单独加LPS比较,培养48 h后,用ELISA法测定培养物上清液中TNF-α浓度。结果与空白培养比较,经LPS诱导活化后COPD组PBMCs培养物上清液TNF-α浓度明显升高(P<0.01),并高于健康对照组(P<0.05);加入LPS+4%喘可治后,COPD组PBMCs培养物上清液TNF-α浓度明显降低(P<0.01)。结论喘可治可以抑制COPD稳定期患者PBMCs分泌的TNF-α,这可能是喘可治治疗COPD的作用机制之一。Objective To explore the effect of Chuankezhi Injection(CKZ) on tumor necrosis factorα(TNF-α) secreted by peripheral blood mononuclear cells(PBMCs) in patients with chronic obstructive pulmonary disease(COPD). Methods PBMCs were isolated from 20 COPD patients at stable phase and 20 healthy control subjects by density gradient method. Some PBMCs were treated with RPMI-1640 media alone, another PBMCs were treated with Lipopolysaccharides(LPS), and the rest were treated with LPS plus 4% CKZ for 48 hours. The concentration of TNF-α in supernate of the culture was detected by ELISA. Results Compared with PBMCs which were treated with RPMI-1640 media alone, the concentration of TNFα in supernate of PBMCs culture which were induced by LPS was increased significantly(P<0.01), and the concentration of TNF-α secreted by PBMCs in COPD group was higher than that in healthy control group(P<0.05). Compared with PBMCs which were treated with LPS, the concentration of TNF-α in supernate of PBMCs culture which were treated with LPS plus 4% CKZ in COPD group was decreased significantly( P<0.01). Conclusion CKZ can inhibit the secretion of TNF-α by PBMCs in COPD patients at stable phase, which may partly explain the mechanism of treating COPD.
