摘 要:采用聚丙烯酰胺凝胶垂直平板电泳(PAGE)技术对草地早熟禾(Poa pratensis L.)和青海扁茎早熟禾(Poa pratensis var.cv.Qinghai)的过氧化物酶(POD)同工酶进行检测和分析,从生化水平探讨两者的亲缘关系及遗传差异。结果表明,草地早熟禾与青海扁茎早熟禾在幼叶期、旗叶期的酶谱分别具共有带8条、6条,其酶带数量、带级、位点及活性强度各不相同,形成了各自特有的酶谱特征。在幼叶期,青海扁茎早熟禾具1条特征谱带,而草地早熟禾没有;在旗叶期,草地早熟禾具3条特征谱带,而青海扁茎早熟禾没有。利用这些酶谱特征找出供试材料间的细微差异是可行的,是两者进行生化水平鉴定的有利指标。The peroxidase isoenzymes(POD) in Poa pratensis L. and Poa pratensis var. cv. Qinghai were detected and analyzed by vertical slab polyacrylamide gel electrophoresis(PAGE) to discuss their genetic relationship and differences on biochemical level. The results showed that the zymogram of Poa pratensis L. and Poa pratensis var. cv. Qinghai during the young leaves and flag leaf stage had eight and six common bands, respectively. But the enzyme strip number, band level, loci and activity were all different and formed specific character of zymogram. The Poa pratensis var. cv. Qinghai had 1 characteristic band C1 at the stage of young leaves, while Poa pratensis L. had not. In the flag leaf stage, Poa pratensis L. had three unique bands, and Poa pratensis var. cv. Qinghai bands had not. It was proved that the zymogram character could be used to find the genetic difference in test materials, it was a favorable index for biochemical detection.
