摘  要:对非转基因罂粟植株进行草甘膦叶喷试验,筛选其最佳叶喷致死浓度;以此浓度草甘膦溶液对T1代转芪合酶基因的罂粟植株进行草甘膦筛选,并对筛选出的阳性植株进行PCR检测。结果表明:罂粟植株草甘膦的最佳筛选浓度为0.01%~0.02%。在转基因罂粟植株3叶一心期和6叶一心期分别叶喷0.01%和0.02%的草甘膦溶液2次筛选后,得到14株转芪合酶基因罂粟阳性植株,经PCR检测得到4株转基因植株,为罂粟抗病种质资源筛选提供了一种新方法。Using non-transgenic poppy plant as material to conduct foliage spray test with glyphosate in order to select the optimal lethal concentration of glyphosate.Then,using the optimal concentration of glyphosate solution to screening T1 generation of stilbene synthase(STS) transgenic poppy plants,and then the selected positive plants were tested by PCR.The results showed that the optimal screening concentration of glyphosate for poppy plant was 0.01%-0.02%.After respectively sprayed 0.01% and 0.02% glyphosate to transgenic poppy seedlings at period of three leaves and one bud and period of six leaves and one bud,total 14 positive poppy plants were obtained,and four STS transgenic poppy plants were confirmed through PCR testing.
