摘  要:目的探讨枸杞多糖(LBP)对高温环境中力竭运动大鼠胸主动脉血管反应性及自由基代谢的影响。方法雄性SD大鼠30只,随机分为对照组、高温力竭运动组、高温力竭运动+LBP组(200mg.kg-1d-1),后两组大鼠分别在高温条件下进行四周游泳训练到力竭。采用离体血管环灌流技术测定大鼠胸主动脉血管环对去甲肾上腺素(NA)的反应性;检测血清中丙二醛(MDA)、超氧化物歧化酶(SOD)、乳酸(LD)、热休克蛋白70(HSP70)以及一氧化氮(NO)的变化。结果与对照组比较,高温力竭运动组胸主动脉血管环对NA的收缩反应增加(P<0.01);其血清中MDA、LD、HSP70含量升高,NO含量降低(P<0.05)。与高温力竭运动组比较,灌服LBP组大鼠能够降低NA对血管环引起的收缩幅度,其血清中SOD、NO及HSP70含量高于高温力竭运动组(P<0.01),而血清中MDA、LD的含量低于高温力竭运动组(P<0.01)。结论 LBP可减弱对NA的血管收缩反应,改善胸主动脉血管弹性,其机制可能与增强血清抗氧化酶活力,减少自由基及脂质过氧化物、提高机体血乳酸耐受力及增加HSP70的表达有关。Objective To evaluate the effect of lycium barbarum polysaccharides(LBP) on the reactivity associated with exercise training of aortic rings and exercise-induced oxidative stress at high temperature in rats.Methods All animals were divided randomly into 4 groups(n=10 each group): control sedentary group(CS);heated exercise group(HE) and heated exercise that received LBP group(HE-LBP).Rats of HE and HE-LBP group were assigned to swim for 4 weeks in a high ambient temperature environment.Concentrations of malondialdehyde(MDA),super oxide dismutase(SOD),lactic acid(LD),nitric oxide(NO) and heat shock protein 70(HSP70) levels were measured.Concentration-response curve to noradrenaline(NA)(10-10~10-5M) was calculated.Results A significantly diminished vasoconstrictor response to NA in aortic rings was found from rats that received LBP group.Meanwhile,LBP increased significantly the activities of SOD,NO and HSP70 levels and attenuated the accumulation of lactic acid and MDA increase(P&lt;0.05).Conclusion LBP can attenuate contractile responses to NA and improve arterial elasticity.
