姜黄素预处理拮抗离体心脏缺血再灌注损伤的作用

摘  要:目的观察姜黄素(Cur)预处理对SD大鼠离体心脏缺血再灌注(IR)损伤是否具有拮抗作用,并研究凋亡相关通路在其中的作用。方法 SD大鼠32只随机分为4组,每组8只,即对照组(Control组)、缺血/再灌注组(IR组)、Cur预处理+IR组(Cur+IR组)、Cur处理组(Cur组)。采用大鼠离体心脏Langendoff逆行灌注模型,Cur预处理时间为5 min,浓度为1μmol/L,缺血45 min,再灌注60 min。分别于缺血前及再灌注后15 min、30 min、45 min和60 min,测定血流动力学各项指标,包括左心室发展压(LVDP)、左心室内压最大变化速率(+dp/dt max)、心率、冠状动脉流量、计算心率压力指数(DP,LVDP×HR/1000)。氯化三苯基四氮唑(TTC)染色法测定各组心肌梗死面积,原位末端转移酶标记技术(TUNEL)测定各组细胞凋亡率,Western blot检测各组抗凋亡相关蛋白Bcl-2和凋亡相关蛋白cytochrome c、Bax的表达。结果与Control组比较,cur组各项指标无统计学差异,再灌注后IR组各时间点血流动力学参数显著降低,心肌梗死面积显著增加,细胞凋亡率升高,Bcl-2表达下降,cytochrome c、Bax表达上升(P<0.01)。与IR组比较,再灌注时Cur+IR组血流动力学显著改善,心肌梗死的面积明显减少,细胞凋亡率降低,Bcl-2表达升高,cytochrome c、Bax表达降低(P<0.01)。结论 Cur预处理有拮抗心肌IR损伤作用,其作用可能与抗凋亡信号通路的活化有关。Objective To investigate the protective effect of curcumin( Cur) pre-treatment against myocardial ischemia reperfusion( IR) injury in isolated rat hearts and explore the possible mechanism involves apoptotic pathway. Methods Thirty-two hearts isolated from SD rats underwent Langendroff perfusion procedures were randomized into 4 groups( n = 8) : Control group,IR group,Cur + IR group and Cur group. Myocardial IR injuries were produced by ischemia for 45 min and followed by 60 min reperfusion. LVDP,± dp / dt max,HR,CF and DP were recorded respectively before ischemia and 15,30,45 and 60 min after reperfusion. The myocardial infarct size was detected with TTC test. The apoptotic index was detected with TUNEL method. The expression of Bcl-2,cytochrome c and Bax was detected with Western blot. Results Compared with the Control group,the hemodynamic parameters in IR group decreased significantly,the infarct size and apoptotic index increased significantly,the cytochrome c and Bax expression increased and the Bcl-2 decreased significantly( P < 0. 05).Compared with the IR group,Cur pre-treatment significantly improved the hemodynamic parameters,decreased the infarct size,apoptotic index,the expression of cytochrome c and Bax,and increased the expression of Bcl-2( P <0. 05). No significant differences were observed in these indicators between the Control group and the Cur group( P> 0. 05). Conclusions Pre-treatment with Cur can significantly reduce myocardial IR injury,and the effect Curcumin pretreatment is closely related to the activation of anti-apoptotic signaling pathway.
