摘 要:目的 观察中国华南地区汉族人群血管紧张素转换酶 (ACE)基因I/D多态性与高血压病(EH)及左室肥厚 (LVH)的相关性。 方法 以 136例原发性高血压病患者和 110例健康体检者为对象 ,测定临床指标 ;经对高血压病患者进行心电图和心脏超声检测 ,分为左室肥厚组 (LVH +) 72例 ,无左室肥厚组 (LVH - ) 6 4例 ;应用多聚酶链式反应 (PCR)、限制性内切酶方法检测ACE基因的多态性分布。 结果 (1)EH组ACE基因I/D多态性与对照组比较基因型和等位基因分布均无显著性差异。 (2 )LVH -组与LVH +组ACE基因I/D多态性基因型和等位基因分布均有显著性差异(P <0 .0 5 ) ,LVH +组Ⅱ基因型和Ⅰ等位基因频率显著高于LVH -组。 结论 (1)ACE基因I/D多态性与中国华南地区汉族人群EH发生无相关性。 (2 )ACE基因Ⅱ多态性与LVH相关。Objective To investigate the relationship of polymorphism of angiotension converting enzyme (ACE) gene with essential hypertension (EH) and left ventricular hypertrophy (LVH) in hypertensive patients. Methods Clinical data were collected in 136 patients with EH and 110 controls. Left ventricular mass and geometry were assessed with ECG and duplex sonography in hypertention patients. DNA was extracted from peripheral blood leukocytes. The insertion/deletion polymorphism of ACE gene was monitored by polymerase chain reaction (PCR). Results The analysis of individual ACE genotypes and allele frequencies distribution showed no significant difference between EH and controls. ACE gene showed a significant difference in genotype or allele frequencies distribution between LVH + groups and LVH - groups (P<0.05). Conclusions No association between the polymorphism of ACE gene and EH is found. A significant association between the polymorphisms of ACE gene in EH is detected, and allele I of ACE gene may be an independently risk factor for LVH in hypertention patients. There is a synergistic effect between the polymorphism of ACE gene and LVH in EH.
