摘 要:目的探讨促红细胞生成素(EPO)对人骨髓间充质干细胞(MSCs)向血管内皮细胞诱导分化的影响。方法从正常人骨髓中分离获取MSCs,体外培养扩增、纯化后,分4组对其进行诱导分化:(1)EPO组;(2)碱性成纤维细胞生长因子(bFGF)+血管内皮细胞生长因子(VEGF)组;(3)EPO+bFGF+VEGF组;(4)DMEM培养液对照组。诱导前后行流式细胞仪检测细胞表面标记CD29、CD31、CD34、CD44,免疫细胞化学鉴定vWF抗体。结果 EPO组、bFGF+VEGF组及EPO+bFGF+VEGF组诱导MSCs后的细胞CD31、CD34表达率升高,CD29、CD44表达率降低,vWF抗体阳性。结论 EPO可以诱导MSCs向血管内皮细胞分化,但联合bFGF、VEGF并不能进一步促进MSCs向血管内皮细胞转化。Objective To investigate the influence of erythropoietin(EPO) on the induction and differentiation of human bone marrow mesenchymal stem cells(hMSCs) into endothelial cells.Methods The hMSCs were isolated from bone marrow of human.After being amplified and purified in vitro,the hMSCs were induced in four groups:(1)EPO group;(2)basic fibroblast growth factor(bFGF)+vascular endothelial growth factor(VEGF)group;(3)EPO+bFGF+VEGF group;(4)DMEM group.Inducted cells were characterized by flow-cytometry CD29,CD31,CD34,CD44 and immunocytochemistry von Willebrand factor(vWF).Results The hMSCs were induced by EPO 、bFGF+ VEGF and EPO+bFGF+VEGF.The expressions of flow-cytometry CD31,CD34 increased and CD29,CD44 decreased.Immunocytochemistry analysis showed typical expression of the vWF.Conclusion The hMSCs can be cultured and differentiated into endothelial cells by EPO in vitro.However,EPO combined with bFGF and VEGF can not further promote the hMSCs to induce into endothelial cells.
