摘 要:目的从健康献血者外周血分离出高纯度、高活性的嗜酸性粒细胞(Eosinophils,EOS)。方法采用免疫磁珠阴性选择法分离EOS,检测其纯度、活性,计算回收率。结果分离得到的EOS细胞形态完整,纯度为(94.8±2.0)%,存活率为(97.5±1.1)%,回收率为(63.0±14.3)%。结论该法能在短时间内(≤4小时)从外周血分离出无细胞损伤、高纯度的EOS,适用于后续EOS相关的实验研究,因此该方法是一种高效快速的分离获取高纯度外周血EOS的免疫学方法。Objectives To isolate eosinophils of high purity and high viability from the peripheral blood of healthy donors.Methods Peripheral blood eosinophils were isolated by immunomagnetic beads negative selection method,their purity and activity and recovery rate were assessed.Results The purity of the EOS isolated by this method reached(94.8±2.0)%,with a survival rate of(97.5±1.1)%and the recovery rate of(63.0±14.3)%.The isolated cells were morphologically intact.Conclusion This procedure applying immunomagnetic beads negative selection method could be accomplished in≤4h and preserves eosinophils in a quiescent,viable state,which are ideal for subsequent experimental research.Therefore,this method is a highly effective immunological method for rapid isolation of high purity of eosinophils.
