摘  要:目的考察采用不同规格的羟丙基甲基纤维素(HPMC)对布洛芬缓释片的体外释药行为的影响,为布洛芬骨架型缓释片的研制提供实验依据。方法选用不同规格的HPMC(K100LV、K4M、K15M、K100M)为骨架材料,将模型药物布洛芬制备成骨架型缓释片,通过测定体外释放度,考察HPMC的黏度、用量以及填充剂种类对布洛芬体外药物释放速率的影响,并对优化处方的释药机制进行探讨。结果 HPMC的黏度、用量以及填充剂的性质对药物释放有显著影响,优化处方的释药过程为药物扩散和骨架溶蚀,是两者共同作用的结果。结论联合应用不同黏度和用量的HPMC,可制备具有理想释药行为的布洛芬骨架缓释片。Objectives To investigate the in vitro release and drug release mechanism of ibuprofen from HPMC matrix tablets. Methods Using ibuprofen as a model drug, HPMC matrix tablets were prepared using wet granulation method and the effect of HPMC viscosity, HPMC content and properties of filler on the in vitro release of ibuprofen from the matrix tablets were studied. Drug release mechanism was discussed. Results The release rate of ibuprofen from HPMC matrix tablets was mainly controlled by theviscosity of HPMC, content of HPMC and properties of the fillers. Drug release mechanism was Fickian diffusion and Erosion. Conclusions Ibuprofen sustained release tablets with ideal release behavior could be prepared by selecting HPMC with different viscosity and adjusting the content of HPMC.
