鼻腔给药地西泮微乳的制备及性质考察

摘　　要：目的制备地西泮微乳并进行质量评价。方法用伪三元相图确定微乳处方,考察其黏度、电导率、表面张力、粒径以及鼻纤毛毒性。结果微乳处方的质量分数组成为:8.0%中链三甘油酯、21.3%Tween80、10.7%PEG400、60.0%水。所得微乳澄清、透明。其黏度为1×10-2Pa·s,电导率为12.3mS·m-1,表面张力为48.5mN·m-1,粒径为31.2nm,并且纤毛毒性低。结论制备了地西泮微乳,为开发地西泮新型鼻腔给药制剂提供了依据。Objective To prepare diazepam microemulsion and evaluate its quality.Methods The formulation of diazepam microemulsion was selected using pseudoternary phase diagram.The viscosity,conductivity,surface tension,droplet size and nasal ciliotoxicity of the microemulsion were characterized.Results The microemulsion was composed of Miglyol 812(8.0 %),Tween 80(21.3 %),PEG400(10.7 %) and water(60.0 %),and its viscosity,conductivity,surface tension and droplet size were 0.010 Pa s,12.3 mS·m-1,48.5 mN·m-1 and 31.2 nm,respectively.The diazepam microemulsion had only slight nasal ciliotoxicity.Conclusion Diazepam microemulsion could be prepared using Miglyol 812-Tween80-PEG400-water system,which can be used for intranasal administration.
