摘  要:目的:探讨裸花紫珠(MDG-1)在血管内皮细胞系统中及鸡胚绒毛尿囊膜模型的促血管生成作用。方法:使用血管生成抗体芯片技术检测MDG-1处理后人微血管内皮细胞中多种血管生长因子的变化情况,以及利用RT-PCR方法检测对碱性成纤维细胞生长因子(bFGF)mRNA表达的影响;并通过观察它对人微血管内皮细胞管腔结构形成的影响,验证其促血管生成效应;同时分别以表皮生长因子(EGF)和生理盐水为阳性和阴性对照,作用于绒毛尿囊膜(CAM),通过一级、二级血管计数,初步评价MDG-1的促血管生成活性。结果:MDG-1对多种血管生长因子都有调节作用,其中对bFGF有明显的上调作用,且并MDG-1可以上调bFGFmRNA表达;MDG-1组管腔数目较生理盐水组明显增多;MDG-1组CAM上血管数比生理盐水组明显增加。结论:MDG-1可以通过上调bFGF而促进人微血管内皮细胞的血管生成以及促进CAM的血管生成。Objective: To investigate the nakedflower beautyberry ( MDG-1 ) in vascular endothelial cell system and model of chick chorioallantoic membrane angiogenesis effects. Methods: The use of angiogenesis antibody chip technology is used in the detection of MDG-1 processing human microvascular endothelial cells in a variety of vascular growth factor changes, and the RT-PCR method is atilized to detect the effects of basic fibroblast growth factor ( bFGF ) on the expression of mRNA; and by observing its effect on human microvascular endothelial cells tube structure formation, its angiogenic generation effect; at the same time respectively epidermal growth factor ( EGF ) and physiological saline as positive and negative controls are compared,the chorioallantoic membrane ( CAM ), through a level, two level of blood vessel counts, preliminary evaluation of MDG-1 angiogenic activity are applied Results: MDG-1 on a variety of angiogenic growth factors has regulating effect on bFGF, which is obvious up-regulated, and MDG-1 can upregulate bFGF mRNA expression; group MDG-1 lumen numbers than the saline group is increased significantly; group MDG-1 CAM number vessels than in the saline group is increased significantly. Conclusion: MDG-1 can promote human microvascular endothelial cell angiogenesis and promote CAM angiogenesis.through the up-regulation of bFGF
