骆驼蓬总生物碱对斜纹夜蛾的拒食活性及生理效应  被引量:5

摘  要:采用酸水总生物碱提取法对骆驼蓬种子乙醇提取物进行初步分离,得到粗品总生物碱。将总生物碱进行柱层析,分离获得16种馏分,4号和8号可析出结晶。生物测定结果表明,以4、12和14号3个馏分的杀虫活性最高,3个馏分1000μg/mL对斜纹夜蛾4龄幼虫24h的拒食率分别为96.04%、92.07%、100.00%;48h的拒食率分别为98.82%、96.05%、98.08%。3种高活性馏分500μg/mL对斜纹夜蛾4龄幼虫的作用机理研究表明,12号和14号馏分能显著减少血淋巴蛋白质含量,药物处理的试虫体内总糖含量明显下降。Total alkaloids extracts were isolated by the primary separation of ethanol extract of the seed of Peganum harmala (ESPH). Separation of chloroform fraction was performed through silica gel column; 16 fractions were isolated; the 4th and 14th fractions produced prisms. Bioactivity of all fractions were compared. The most active fraction were fraction 4, 12 and 14. The bioactivity test showed that fraction 4, 12 and 14 at 1 000 μg/mL possessed very high strong antifeedant property against the 3rd-instar larvae of Spodoptera litura in no-choice test, and the antifeedant rates were 96.04%, 92.07% and 100.00% after 24 h, and 98.82%, 96.05% and 98.08% after 48 h, respectively. Mechanism of three active fractions showed that active substance of P. harmala could disturb normal physiological function of insect. Against the 4th-instar larvae of S. litura, the protein content of haemolymph was significantly reduced in the larvae, and the total body sugar was reduced obviously when treated with the 12th and 14th fractions at 500 μg/mL.
