摘 要:目的观察不同剂量益气化瘀化痰养阴方药对肝纤维化大鼠肝组织结构及过氧化物酶体增殖物激活受体(PPARγ)表达的影响。方法将雄性SD大鼠随机分为正常组、肝纤维化模型组、秋水仙碱组、益气化瘀化痰养阴低剂量组、中剂量组及高剂量组。除正常组外,其余各组均采用"皮下注射CCl4+低蛋白高脂饮食+酒精饮料"的造模方法制备肝纤维化大鼠模型。正常组和模型组灌服生理盐水,其余各组分别灌服秋水仙碱及不同剂量的益气化瘀化痰养阴中药煎剂。12 w后处死大鼠,通过光镜和电镜观察各组肝脏病理学变化,并采用RT-PCR及Western印迹方法检测各组大鼠肝组织PPARγ基因和蛋白表达水平。结果与正常组相比,模型组大鼠肝组织PPARγ基因和蛋白表达明显减少(P<0.05);与模型组相比,各治疗组肝组织内纤维化程度均有不同程度减轻;秋水仙碱组、益气化瘀化痰养阴中剂量及高剂量组大鼠肝组织PPARγ基因和蛋白表达明显增加(P<0.05),其中以益气化瘀化痰养阴中、高剂量组最明显;益气化瘀化痰养阴中剂量组及高剂量组间无明显差异(P>0.05)。结论中、高剂量益气化瘀化痰养阴中药可以减轻肝纤维化模型大鼠肝组织内纤维化程度,其机制可能与增加肝组织内PPARγ的表达有关。Objective To study the effect of differentdosage of Yiqi Huayu Huatan Yangyin Formula Decoction on liver structure and expression of peroxisome proliferator-activated receptor γ( PPARγ) in rats with liver fibrosis .Methods 110 male healthy SD rats were ran-domly allocated into control (n=10), liver fibrosis (n=20), colchicine (n=20), YHHY-low-dosage-treated (n=20),YHHY-middle-dos-age-treated (n=20), YHHY-high-dosage-treated (n=20) groups.Rats in control group were fed by normal food and water ,while the others were prepared of liver fibrosis by the methods of “subcutaneous injection of carbon tetrachloride (CCl4) +low-protein and high-fat diet+liq-uor”.After 12 weeks of treatment , liver tissue histological examination was carried out to examine the liver pathology .The expression levels of PPARγmRNA and protein were measured by RT-PCR and Western blot .Results In contrast with control group , significant fibrosis and PPARγmRNA and protein expressions of the liver tissue were decreased significantly on liver fibrosis group (P<0.05).Compared with liver fibrosis group, liver fibrosis was inhibited in colchicine, YHHY-middle-dosage-treated, YHHY-high-dosage-treated groups (P<0.05). However ,no statistically significant differences were found between YHHY-middle-dosage-treated and YHHY-high-dosage-treated groups ( P>0.05 ) .Conclusions Middle-dosage or high-dosage of YHHY could attenuate liver fibrogenesis in rats .Its effective mechanism might be related to increase PPARγmRNA and protein expressions in liver tissue .
