摘　　要：目的：通过改变小鼠corin蛋白结构来研究跨膜区和胞外段对corin酶原激活和质膜锚定的影响。方法采用定点突变的方法构建小鼠和人corin蛋白跨膜区和胞外段互换的突变质粒，转染HEK293细胞后，Western blotting检测corin表达及活化情况，流式细胞术检测corin在细胞膜上表达情况。结果发现小鼠corin蛋白的跨膜区换成人corin跨膜区之后，并未影响corin蛋白表达、活化及质膜锚定；但是人corin胞外段换成小鼠的之后，则可以促进corin蛋白活化及质膜锚定。结论小鼠corin胞外段存在可能促进corin蛋白酶原活化及质膜锚定的序列。Objective Functional analysis of mouse corin zymogen activation and cell surface targeting by changing the transmembrane and extracellular domain. Methods We constructed mutant plasmids by QuickChange site-directed mutagenesis. After HEK293 cells were transfected, expression and activation of corin was detected by Western blotting, and cell surface targeting of corin was measured using flow cytometry. Results The transmembrane domain is not necessary for the biological activity of mouse corin, and the extracellular domain of mouse corin promote the activation and cell surface targeting of human corin. Conclusion There may exist motif in mouse corin extracellular domain which can promote corin zymogen activation and membrane anchoring.
