摘　　要：目的探讨益气养阴、消牴通络中药对早期糖尿病大鼠肾组织Podocin基因表达的影响。方法将SD大鼠随机分为6组:肾切组、模型组、厄贝沙坦组、中药低、中、高剂量组,按各组相应药物剂量灌胃。于第6周末检测各组大鼠24 h尿蛋白(UPro)量及肾功能水平;RT-PCR检测肾皮质Podocin表达水平。结果厄贝沙坦、中药低、中剂量组与模型组比较UPro量显著下降(P<0.05);各治疗组与模型组比较,肾功能明显改善(P<0.05),且Podocin表达显著增加(P<0.01)。结论益气养阴、消牴通络中药可能通过减少Podocin蛋白的丢失,使肾小球滤过膜的完整性得到一定程度的恢复而减轻蛋白尿,从而发挥其对糖尿病肾病大鼠的治疗作用。Objective To explore the influence of supplementing Qi-nourishing Yin-dispersing Blood Stasis-Dredging Collateral Herbsongene expression of Podocin in early diabetes rats kidney organization.Methods Model rats randomly were divided into 6 groups:simple nephrectomy model, irbesartan,Traditional Chinese Medicine (TCM) low,middle and high dose groups.The rats were orally adminis-tered with corresponding dose of drug.At the end of 6th week, 24 h urine protein (UPro), serumcreatinine (Scr) and urea nitrogen (BUN) level were detected.Expression of Podocin in renal cortical was measured with RT-PCR.Results UPro level of irbesartan group ,TCM low and middle dose groups were decreased significantly (P<0.05),compared with those of the model group ,Scr and BUN levels of the model group and BUN level of the treated groups were increased ( P<0.01 ).Conclusions Supplementing Qi-nourishing Yin-dispersing blood Sta-sis-dredging collateralherbs could protect the glomerular filtration barrier hole in the outer membrane and podocyte structure and function ,re-duce protein uria and delay the development of diabetic nephropathy.
