摘 要:目的观察母鼠在孕期和哺乳期壬基酚(NP)暴露对雄性仔鼠肾上腺TNF-α和IL-1β表达水平的影响及血清中激素水平的变化。方法根据妊娠时间将32只孕鼠随机分为对照组及低、中、高剂量暴露组;消化道暴露,暴露剂量分别为0、25、50、100 mg/(kg·d);暴露时间为受孕第6 d到出生后21 d(仔鼠哺乳结束)。免疫组织化学法检测仔鼠出生后21、60 d肾上腺TNF-α和IL-1β表达;放免法检测血清中激素水平的变化,并分析上述变化与NP暴露之间的关系。结果与对照组比较,低、中、高剂量组21、60 d TNF-α和IL-1β光密度增高(F=4265.850;96.191及428.759;1519.815,P<0.05),随给药剂量和给药时间的增加,21、60 d时的光密度增高,与NP暴露剂量呈正相关。血清睾酮水平随NP暴露剂量增加而降低,雌二醇水平随NP暴露剂量的增加而增加。结论 NP孕期暴露使仔鼠肾上腺炎性介质TNF-α和IL-1β表达增加,炎症性反应可能是NP引起仔鼠肾上腺损伤的机制,而肾上腺的损伤可能导致肾上腺功能异常并进而影响仔鼠的血清激素水平。Objectives To observe the adverse effects of nonylphenol( NP) exposure during pregnancy and lactation on TNF-α and IL-1β in the adrenal glands of pups and the change of their serum hormone levels.Methods Thirty two pregnant rats were randomly divided according to the time of conception into a control group and three exposure groups( low,middle and high dose group),and were exposed to NP through digestive tract at the dose of 0,25,50 and 100 mg /( kg·d),respectively,started from the 6thday after conception and ended at the21thof stopping lactation. The expression of TNF-α and IL-1β in the adrenal gland of rat pups at 21d and 60d after birth was detected by immunohistochemical method,the association between NP exposure with the expression of TNF-α and IL-1β was analyzed,and the change of serum hormone level was also observed. Results In comparison with the offspring in control group,the integrated optical density( IOD) for TNF-α and IL-1β in the adrenal gland of 3 exposure groups at 21d and 60d after birth were higher( F = 44265. 850; 96. 191and 428. 759;1519. 815,P < 0. 05). The dose and time of NP exposure were associated with the expression of TNF-α and IL-1βin a dose-response manner. The levels of serum testosterone decreased with higher dose of NP exposure,and estradiol increased with higher dose of NP exposure. Conclusions Exposure to NP during gestation might induce an increased expression of inflammatory mediators( TNF-α,IL-1β) in the adrenal glands of pup rats. Inflammatory reaction might have an injury mechanism on the adrenal glands of pup rats exposed to NP during gestation. The injury of adrenal gland might lead to adrenal dysfunction and,in turn,affect the serum hormone level in pups.
