摘  要:目的探讨羟基红花黄色素A防治激素性股骨头缺血坏死的作用机理。方法用大剂量地塞米松诱导体外培养家兔骨髓间充质干细胞成脂分化,在诱导其成脂分化的同时给予不同浓度的羟基红花黄色素A进行干预,干预5 d后用蛋白免疫印迹方法测定各组细胞ERK1和ERK2蛋白的表达。结果羟基红花黄色素A干预组细胞ERK1和ERK2蛋白的表达较对照组明显升高,差异具有统计学意义(P<0.05)。结论羟基红花黄色素A防治激素性股骨头缺血性坏死的作用机理不仅是改善坏死股骨头局部血运,同时还与其促进骨髓间充质干细胞向骨细胞转化、增殖有关。Objective To investigate the therapeutic mechanism of steroid-induced avascular necrosis of femoral head treated by HSYA. Methods Adipogenic differentiation of BMSCs cultured in vitro was induced by heavy doses of steroid, and HSYA with different concentrations were added into the culture system simultaneously. After 5 days, the protein expressions of ERK1 and ERK2 were detected by Western Blotting. Results Compared with the control group, the protein expressions of ERK1, ERK2 in HSYA grous with different doses were increased respectively(P<0.05). Conclusion HSYA has good effects on preventing and treating steroid induced avascular necrosis of femoral head, and its mechanism is not only related with the ef-fect of improving local blood circulation of necrotic femoral head, but also related with the effect of promoting BMSCs proliferation and differentiation into osteocytes.
