摘 要:目的观察透骨消痛胶囊含药血清对体外培养大鼠退变软骨细胞表达caveolin-1、p38MAPK的影响,探讨透骨消痛胶囊防治骨性关节炎的作用机制。方法 4周龄SD大鼠膝关节软骨建立体外培养的软骨细胞,采用Ⅱ型胶原原位杂交法对第3代软骨细胞进行鉴定。用10 ng/mL IL-1β干预第3代软骨细胞复制退变软骨细胞模型。模型复制成功后,随机分为模型组和透骨消痛胶囊含药血清组,分别在培养24 h、48 h、72 h后用TaqMan real-time PCR和Western Blot法检测各组mRNA和蛋白的表达。结果透骨消痛胶囊含药血清组caveolin-1和p38MAPK基因、蛋白相对表达量随着干预时间的延长逐渐下降,统计均有显著性差异(P<0.05)。结论透骨消痛胶囊治疗骨性关节炎的部分作用机制是有效抑制退变软骨细胞caveolin-1、p38MAPK的表达。Objective To observe the effects of serum containing Tougu Xiaotong capsule on the expression of caveolin-1 and p38MAPK in rats degenerated chondrocytes cultured in vitro, and explore it 's mechanism of treating osteoarthritis. Methods Chondrocytes were derived from the cartilages of knee joints of 4-week-old Sprague-Dawley rats, and the passage 3rd chondrocytes were identified with in situ hybridiza-tion of type Ⅱ collagen. The passage 3rd chondrocytes were treated with 10 ng·mL-1 IL-1β to copy model of degenerated chondrocytes, then the chondrocytes were divided into model group and serum containing Tougu Xiaotong capsule group, and intervened by corresponding culture solution for 24 h, 48 h and 72 h respective-ly. The mRNA and protein expression of caveolin-1 and p38MAPK in each group were detected by TaqMan real-time PCR and Western Blot. Results In serum containing Tougu Xiaotong capsule group, the mRNA and protein relative expression of caveolin-1 and p38MAPK were gradually decreased with the prolongation of intervention time, and there was a significant difference in the mRNA and protein expression between the two groups at different time points(P<0.05). Conclusion Tougu Xiaotong capsule can significantly inhibit the expression of caveolin-1 and p38MAPK in degenerated chondrocytes, which may partly explain the mechanism of treating osteoarthritis.
