摘  要:目的观察筋骨草醇提物抗小鼠肺纤维化的作用机制。方法 72只小鼠随机分6组,分别为假手术对照组、肺纤维化模型对照组、醋酸泼尼松组、筋骨草醇提物高、中、低3个剂量组(12、6、3 g/kg/d),每组12只。除假手术对照组外,其余各组采用气管内注入博莱霉素方法复制小鼠肺纤维化模型,假手术对照组小鼠气管滴注等体积生理盐水,于造模当天开始每日给药,分别于第7、28 d处死,观测小鼠肺组织肺泡炎和纤维化程度,检测肺组织转化生长因子-β(TGF-β)的表达和血清白介素-4(IL-4)含量。结果筋骨草醇提物高、中剂量组治疗后小鼠肺泡炎及肺纤维化较模型组显著减轻(P<0.01,P<0.05);筋骨草醇提物高、中、低3个剂量组的TGF-β和IL-4含量显著低于模型组(P<0.01,P<0.05),呈量效关系。结论高、中剂量筋骨草醇提物可通过下调细胞因子TGF-β和IL-4的表达达到减轻小鼠肺间质纤维化的效果。Objective To explore the mechanism of ethanol extract of Ajuga Decumbens(EEAD) in treating experimental pulmonary fibrosis(PF). Methods 72 mice were randomly divided into sham operation group, PF model group, Prednisone Acetate group and EEAD groups with different doses, and each group had 12 mice. Except sham operation group, other groups were treated with intratracheal instillation of bleomycins to copy PF mice model, sham operation group received same volume of normal saline, and each group received corresponding drugs simultaneously. Mice in each group were executed at the 7th and 28th day, airsacculitis of lung tissue and degree of fibration were observed, and the expression of TGF-β in lung tissue and level of serum IL-4 were detected. Results After the treatment, the degree of airsacculitis and PF in middle, high dosage of EEAD group was lightened respectively compared with the model group(P<0.01, P<0.05). The levels of TGF-β and IL-4 in EEAD groups with different doses were lower respectively than that of model group(P<0.01, P<0.05), which showed a dose-effect relationship. Conclusion Middle and high dosage of EEAD could significantly alleviate pulmonary interstitial fibrosis of mice by down-regulating the expression of TGF-β and IL-4.
