摘  要:目的观察补肾活血方对血管性认知障碍模型大鼠兴奋性氨基酸(EAA)与抑制性氨基酸(IAA)的影响。方法 3月龄Wistar大鼠用双侧颈总动脉永久结扎法制作认知障碍模型,用补肾活血方水煎剂和银杏叶片水溶剂灌胃14 d,观察造模前后大鼠行为的改变,用HPLC法检测大鼠海马组织乙酰胆碱(Ach)、5-羟色胺(5-HT)、谷氨酸、γ-氨基丁酸含量,WB、RT-PCR法检测其受体表达。结果补肾活血方组与模型组比较,逃避潜伏期缩短,翻越旷场实验箱格数增加,海马组织Ach、5-HT、GABA含量及其受体的表达增高,Glu的含量及其受体的表达降低。结论补肾活血方有保护大脑学习记忆能力,降低脑内Ach、5-Ht含量的功效,其作用机制与调节脑内EAA和IAA含量的平衡有关。Objective To observe the effects of Bushen Huoxue decoction on levels of excitatory amino acids(EAA) and inhibitory amino acids(IAA) in rat models of vascular cognitive impairment. Methods 3-month-old Wistar rats were used to establish models of cognitive disorder by bilateral common carotid arteries ligation, then received Bushen Huoxue decoction and hydrosolvent of Ginkgo Biloba Leaves Extract Tablets by gastric perfusion for 14 days. The behavior changes of rats before and after creation of models were observed, the levels of acetylcholine(ACh), 5-HT, glutamic acid(Glu) and gamma-aminobutyric acid(GABA)were detected by HPLC, and the acceptor expressions were detected by RT-PCR and Western Blot. Results Compared with the model group, the escape latency in Bushen Huoxue decoction group was shortened, the open-field test score was increased, the levels of ACh, 5-Ht, GABA and their acceptor expressions were increased, and the level of Glu and its acceptor expression was decreased. Conclusion Bushen Huoxue decoction has protective effect on learning and memory ability of brain, and decreases the levels of ACh and 5-Ht. The mechanism may be related with regulating the balance of EAA and IAA levels.
