摘 要:目的观察中风Ⅱ号对急性脑缺血大鼠脑组织血清肿瘤坏死因子-α(TNF-α)、白细胞介素—1β(IL-1β)含量的影响。方法将60只SD大鼠随机分为正常对照组、缺血组、中风II号低剂量组、中风II号高剂量组,线栓法制备大脑中动脉缺血(MCAO)模型,用双抗体夹心ELISA法测定大鼠血清TNF-α、IL-1β含量。结果中风II号低剂量组、高剂量组均能明显改善MCAO大鼠神经功能缺损评分,降低血清TNF-α、IL-1β含量,高剂量组作用更为明显。结论中风II号可通过抑制促炎细胞因子TNF-α、IL-1β的过高表达,抑制炎症级联反应,减轻急性脑缺血大鼠的炎症损伤。Objective To observe the effect of No.2 prescription for stroke on levels of serum tumor necrosis factor(TNF-α) and Interleukin-1beta(IL-1β) in brain tissue of rats with acute cerebral ischemia.Methods 60 SD rats were randomly divided into normal control group, ischemic group, stroke-Ⅱ low-dose group and stroke-Ⅱ high-dose group. Cerebral ischemia model of middle cerebral artery occlusion(MCAO)was established in rats by using suture method, and the levels of serum TNF-α and IL-1β in rats were detected by double antibody sandwich ELISA method. Results The score of neurologic impairment of MCAO rats in stroke- Ⅱ low-dose group and stroke- Ⅱ high-dose group was significantly improved, the levels of serum TNF-α and IL-1β were decreased respectively, and the effect of stroke-Ⅱ high-dose group was more significant. Conclusion No.2 prescription for stroke can reduce inflammatory injury in rats with acute cerebral ischemia by inhibiting the high expression of proinflammatory cytokines TNF-α and IL-1β and the inflammatory cascade reaction.
