摘 要:旨在分析马波沙星在猪体内的残留消除规律并为制定休药期提供依据。30头健康猪按体重以2mg·kg-1肌注给药,每天1次,连续给药3d,最后一次给药后第0.5、1、3、5、8天随机宰杀5头猪取样。试样用三氯甲烷提取,45℃下氮气吹干和正己烷净化后,经高效液相色谱C18色谱柱分离后用紫外检测器检测。结果显示:给药后12h肾组织中的药物浓度最高,为4.93μg·g-1,第5天所有组织的药物浓度均低于最高残留限量。采用Winnonlin软件计算各组织的消除动力学参数,消除快慢依次为肌肉、注射位点、肾、肝和皮脂,其消除半衰期(t1/2β)分别为17.86、17.92、18.20、18.90、21.20h。肾的药时曲线下面积(AUC)最高,为200.16μg·h·g-1,说明肾是马波沙星在猪体内作用的靶器官。参照EMEA对马波沙星制定的MRL,计算得肾的休药期最长,为5.46d。马波沙星注射液在猪体内吸收迅速,分布广泛,消除缓慢,建议休药期为6d。This experiment was conducted to study tissue residues of marbofloxacin in pigs,preparing for determination of the withdrawal time.Marbofloxacin was administered to 30healthy pigs intramuscularly at a dosage of 2mg·kg-1 for 3consecutive days.Animals were sacrificed at 0.5,1,3,5,8dafter administration randomly.The tissues were extracted with chloroform,dried under nitrogen at 45℃and cleaned up with hexane.The analytes was detected by UV absorptive spectroscopy after separation by C18column.The results showed that kidney had the highest drug concentration with 4.93μg·g-1 at 12h,the concentration of marbofloxacin in all of tissues at 5d was lower than the Maximum Residue Limit(MRL).The pharmacokinetic parameters were estimated using Winnonlin software package.The results indicated that the elimination rate was muscle>injection site>kidney>liver>skin/fat,with the elimination half-life of 17.86,17.92,18.20, 18.90,21.20h,respectively.Compared with other tissues,the area under concentration-time curve(AUC)in kidney was highest with 200.16μg·h·g-1.Kidney was considered as the target organ.According to the MRL set by EMEA,the longest withdrawal time was suggested in kidney with 5.46d.The results demonstrated that absorption of marbofloxacin in Pigs after Intramuscular Administration was quickly,distribution was wide and elimination was slowly.It is proposed that the withdrawal time was 6days.
