3种中药复方对脑缺血再灌注大鼠脑组织细胞间黏附分子-1和血管内皮生子因子表达的影响

摘　　要：目的比较3种不同中药复方对大鼠脑缺血再灌注大鼠脑组织细胞间黏附分子(ICAM)-1和血管内皮生长因子(VEGF)蛋白表达的动态影响。方法采用线栓法制作大鼠局灶性脑缺血再灌注模型,随机分为假手术组、模型组、益气活血方组、镇肝息风汤组和星蒌承气汤组,分别给予相应药物灌胃;采用免疫组化SABC染色法分别检测缺血半暗带ICAM-1和VEGF蛋白表达。结果模型组再灌注各时间点缺血半暗带ICAM-1和VEGF蛋白表达显著升高;各治疗组再灌注各时间点缺血半暗带ICAM-1显著降低和VEGF蛋白表达显著升高,24 h在降低ICAM-1和在升高VEGF方面均以镇肝熄风汤为优;48 h在降低ICAM-1方面以星蒌承气汤组为优,在升高VEGF方面以潜阳熄风组为优;72 h在升高VEGF方面以星蒌承气汤为优。结论 3种中药复方可通过抑制ICAM-1蛋白表达,升高VEGF蛋白表达保护血管内皮对抗脑缺血损伤。Objective To compare with the influence of three Chinese herbal compound formula on the expression of ICAM-1 and VEGF protein in cerebral tissue after focal ischemia-reperfusion( I / R) in rats,and investigate the mechanism of those in resisting on local cerebral I / R inflammatory injury. Methods The local cerebral I / R model was established by ligation of the middle cerebral arteries. The animals were divided into sham-operative,model,the Yiqihuoxue recipe( YHR),the Zhengganxifeng decoction( ZXD) and the Xinglouchengqi decoction( XCD) groups. ICAM-1 and VEGF protein expression were detected by immunohistochemisty method in ischemic penumbra. Results The positive cell number of ICAM-1 and the positive cell of VEGF were higher in model group. The positive cell number of ICAM-1 was lower in model group than that of sham-operative group at all time point after I / R,and the positive cell of VEGF was higher in model group than that of sham-operative group at all time point after I / R. Compared with model group,the positive cell number of ICAM-1 was lower in all treatment groups,the positive cell of VEGF was higher in all treatment groups. The optimal effect was shown in ZXD group( ICAM-1 and( VEGF) at 24 h,in XCD group( ICAM-1) and in ZXD group VEGF) at 48 h,and in XCD groups( VEGF) at 72 h after I/ R. Conclusions The different traditional Chinese medicinal compounds could reduce pathologic injury after focal cerebral I / R in rats by decreasing the expression of ICAM-1 protein and increasing expression of VEGF protein.
