摘 要:目的研究开口箭不同提取部位对异丙肾上腺素(ISO)诱导的小鼠心肌肥厚的影响及其作用机制。方法采用皮下注射ISO致小鼠心肌肥厚模型,分别用开口箭不同提取部位和卡托普利灌胃给药,每天1次,连续14 d。于末次给药的次日进行实验,计算小鼠全心重量指数和左心重量指数,观察心肌病理学改变;测定心肌组织中羟脯氨酸(Hyp)含量、超氧化物歧化酶(SOD)、谷胱甘肽过氧物酶(GSH-Px)活性,丙二醛(MDA)含量,一氧化氮合酶(NOS)活性和一氧化氮(NO)含量,心肌细胞直径和横截面积、心肌间质胶原容积分数。结果与正常组相比,模型组小鼠心脏重量指数、心肌组织中iNOS活性和MDA、Hyp、NO含量明显升高,SOD、GSH-Px和cNOS活性显著降低,心肌细胞直径和横截面积、心肌间质胶原容积分数明显增加(均P<0.01)。经开口箭4个不同提取部位治疗后,上述异常的指标得到了明显改善,与模型组比较,开口箭正丁醇和乙酸乙酯部位均具有显著性差异(P<0.05和P<0.01),其作用效果与卡托普利相当;石油醚部位在改善SOD、Hyp、cNOS、NO和心肌横截面积、心肌间质胶原容积分数等方面有显著性差异(P<0.05);水部位除GSH-Px、iNOS和心肌细胞直径外,均具有显著性差异(P<0.05)。结论开口箭不同提取部位对ISO致小鼠心肌肥厚具有较强的保护作用;在开口箭的4个提取部位中,其作用效果以正丁醇部位最为显著,其余依次为乙酸乙酯部位、水部位和石油醚部位。实验结果表明,清除自由基、抗脂质过氧化及逆转NOS系统异常的表达水平,提高机体的抗氧化能力可能是其抗心肌肥厚的作用机制之一。Objective To investigate the effects of different fractions extracted from Tupistra chinensis Bak. on isoproterenol( ISO)-induced myocardial hypertrophy in mice and its mechanism. Methods ISO was given subcutaneously to induce myocardial hypertrophy in mice. Therapeutic groups were given different fractions extracted from Tupistra chinensis Bak. and captopril after ISO treatment. After 2 weeks intervention,the myocardial hypertrophy indexes were expressed as heart weight / body weight( HW / BW) and left ventricle weight / body weight( LVW / BW),the histological changes were investigated by HE. SOD,GSH-Px,cNOS,iNOS activities and hydroxyproline( Hyp),MDA,NO contents and the cardiomyocyte diameter,the cross section area of cardiomyocyte,collagen volume fraction in the myocardial tissue were assayed,respectively. Results Compared with the measurements in the control group,the myocardial hypertrophy indexes of HW / BW and LVW / BW,the collagen volume fraction,the iNOS activity and MDA,Hyp,NO contents in the myocardial tissue were significantly increased,the activities of SOD,GSH-Px and cNOS were remarkably decreased in model group( P<0. 01,respectively); the cardiomyocyte diameter,the cross section area of cardiomyocyte,collagen volume fraction were also significantly increased( P<0. 01,respectively). When treated with different fractions extracted from Tupistra chinensis Bak,the upper abnormal results were significantly improved. The effects of the n-butyl alcohol and ethyl acetate fractions were significant as compared with those of model group( P<0. 05 and P<0. 01,respectively),and the effect of treatment with captopril quite. Petroleum ether fraction revealed a statistically significant in improving SOD,Hyp,cNOS,NO levels and the cross section area of cardiomyocyte,collagen volume fraction compared with those of model group( P<0. 05,respectively),and that of water fraction was also significant as compared with the model group except for in the case of GSH-Px, iNOS and the cardiomyocyte diameter( P<0. 05,respectiv
