摘 要:目的通过离体细胞实验研究消癥活络方药含药血清对转化生长因子(TGF)-β1诱导的人肾近曲小管上皮细胞(HK-2)转分化的影响。方法体外培养人肾HK-2,应用不同浓度消癥活络方药含药血清处理经TGF-β1诱导活化的HK-2细胞,噻唑兰(MTT)法检测细胞增殖,免疫组化法检测α-平滑肌肌动蛋白(α-SMA)及结缔组织生长因子(CTGF)的表达。结果 TGF-β1可刺激HK-2细胞增殖,消癥活络方药含药血清可明显抑制TGF-β1引起的HK-2细胞增殖。TGF-β1可引起HK-2细胞α-SMA及CTGF表达上调,消癥活络方药含药血清可抑制其上调。结论消癥活络方药对TGF-β1诱导的HK-2的增殖及α-SMA、CTGF的表达具有抑制作用,可拮抗TGF-β1诱导的HK-2细胞转分化作用,抑制TGF-β1刺激HK-2分泌细胞外基质可能是其抗纤维化形成的机制之一。Objective To observe the effect of xiaozhenghuoluo prescription on the transdifferentiation of human renal tubular epithelial cells( HK-2) induced by TGF-β1in vitro. Methods Human renal tubular epithelial cells( HK-2) activated by TGF-β1were treated with the medicated serum of different concentrations. The expressions of α-SMA and CTGF in the cells were detected by immunohistochemistry,and the proliferation and inhibition rate of HK-2 cells were examined by MTT. Results HK-2 was induced proliferation by TGF-β1. Xiaozhenghuoluo prescription significantly inhibited HK-2 proliferation. The expressions of α-SMA and CTGF were increased in HK- 2 induced by TGF-β1,and was down regulated expression in xiaozhenghuoluo prescription groups compared with that of control group. Conclusions Xiaozhenghuoluo prescription could inhibit TGFβ1induced proliferation and the expressions of α-SMA,CTGF in HK-2,one of anti-fibrosis mechanisms might be correlated with inhibiting transdifferentiation of HK-2.
