摘 要:目的观察人参皂甙对氯胺酮麻醉后老年大鼠学习和记忆的影响。方法将48只老年大鼠随机分为4组,每组12只:对照组(A组)、氯胺酮组(B组)、人参皂甙低剂量组(C组)、人参皂甙高剂量组(D组)。用Morris水迷宫检测人参皂甙对氯胺酮所致学习记忆障碍的影响,同时采用ELISA方法检测大鼠海马区脑源性神经营养因子(BDNF)和神经生长因子(NGF)的含量,利用蛋白免疫印迹法测定海马区ERK和CREB的磷酸化水平。结果人参皂甙可改善氯胺酮麻醉后老年大鼠的学习和记忆能力;与氯胺酮组相比,人参皂甙治疗组海马区BDNF、EGF、p-ERK1/2和p-Ser133-CREB的水平均显著增加。结论人参皂甙可明显改善氯胺酮麻醉后老年大鼠的认知功能障碍,其机制可能与其上调ERK和CREB的磷酸化水平并增加BDNF的表达有关。人参皂甙可用于老年人麻醉术后认知功能障碍的治疗与防护。Objective To examine the effects of panax ginsenosides on dysfunction of learning-memory ability in aged mice after ketamine anesthesia,and explore its molecular mechanism. Methods The aged mice were divided randomly into 4 groups: control( A group),ketamine-injected( B group),low( C group),high dose of panax ginsenosides-treated groups( D group). Praxiology of impaired memory acquiring mice caused by ketamine was investigated with Morris Water Maze. The levels of nerve growth factor( NGF) and brain-derived neurotrophic factor( BDNF) in hippocampus of aged rats were examined by ELISA. The phosphorylation of extracellular signal-regulated kinase( ERK) and cAMP-response element binding protein( CREB) in hippocampus were also investigated by Western blot. Results Panax ginsenosides prevented the decline of learning-memory ability in ketamine-treated aged mice. Compared with B group,the levels of NGF,BDNF,p-ERK1 /2 and p-Ser133-CREB in hippocampus of C and D group were significantly increased. Conclusions Panax ginsenosides could improve the cognitive dysfunction in aged rats after ketamine treatment. The molecular mechanism underlying these protective effects is that panax ginsenosides increase the levels of NGF and BDNF through activating ERK-CREB pathway. Panax ginsenosides might be used in the treatment on postoperative cognitive dysfunction in aged people.
