摘 要:目的研究金香丹对缺血再灌注大鼠心肌细胞凋亡的影响。方法采用在体结扎大鼠冠状动脉左前降支建立心肌缺血再灌注模型,采用细胞凋亡测试盒(TUNEL)法观察心肌细胞凋亡情况,免疫组化法检测凋亡相关基因Bax、Bcl-2和Caspsase-3表达的变化。结果模型组细胞凋亡指数、Bax、Bcl-2及Caspsase-3的蛋白表达显著高于假手术组,金香丹组细胞凋亡指数、Bax及Caspsase-3的蛋白表达均显著低于模型组,Bcl-2的蛋白表达显著高于模型组。结论金香丹可以抑制缺血再灌注心肌细胞的凋亡,其作用可能与上调Bcl-2蛋白的表达同时下调Bax、Casperse-3蛋白的表达有关。Objective To explore study the effects of Jinxiangdan on the apoptosis in rats with myocardial ischemia-reperfusion injury. Methods Rats models with myocardial ischemia-reperfusion injury were established by occluding left coronary artery( LCA) and followed by release of it with ligation. TUNEL and S-P immunohistochemistry staining were used to determine the apoptosis and protein expression of Bax,Bcl-2 and Caspase-3 genes. Results The protein expressions of Bax,Bcl-2 and Caspase-3 were increased in the ischemiareperfusion group. The protein expressions of Bax and Caspase-3 were decreased,Bcl-2 was increased significantly in the Jinxiangdan group, compared with those of the ischemia-reperfusion group. Conclusions Jinxiangdan has a good antagonist action on the apoptosis of ischemia / reperfusion myocardial cells,and its mechanisms maybe by increasing the expression of Bcl-2 as well as reducing the expression of Bax and Caspase-3.
