针刺治疗对COPD模型大鼠Th1/Th2细胞亚群格局的影响

摘　　要：目的观察慢性阻塞性肺疾病(COPD)大鼠经针刺治疗后外周干扰素-γ(IFN-γ)和白细胞介素-4(IL-4)的变化,初步探讨针刺对辅助淋巴细胞(T cell helper,Th)亚群Th1/Th2的影响。方法 SD大鼠分为4组:对照组、模型组、针刺7次和14次组,香烟和脂多糖复合方法复制COPD大鼠模型,电针治疗后,观察肺功能和病理学变化,用ELISA方法对大鼠支气管肺泡灌洗液(BALF)及血浆中IFN-γ、IL-4含量进行检测。结果针刺治疗后大鼠的吸气和呼气阻力均比模型组显著下降,肺动态顺应性增加(均P<0.01),且肺组织的炎症病理表现有改善;模型组大鼠BALF及血浆中IFN-γ、IL-4含量与对照组相比均有显著升高(P<0.01或P<0.001),针刺后,均有不同程度下调(P<0.05或P<0.001)。结论针刺治疗可以减轻COPD模型大鼠病理特征和改善肺通气功能,其免疫机制可能与针刺能下调IFN-γ和IL-4,使Th1/Th2格局趋于平衡有关。Objective To observe the change of INF-γand IL-4 in periphery,and deplore acupuncture effect on T cell helper Th1/Th2 cell subsets in chronic obstructive pulmonary disease(COPD) rats after electro-acupuncture(EA) therapy.Methods 40 Spraguedawley rats were randomly divided into control,COPD,EA 7 and 14 times groups.COPD model rats were treated with cigarette smoke and lipopolysaccharide(LPS) for 12 w.EA treatment,begun on the 9th week,was given every other day for 1 w and 2 w continuously.The COPD index of lung pathology and function were observed.The IFN-γand IL-4 levels were performed by ELISA to quantitate the Th1/Th2 cell subsets in peripheral plasma and bronchoalveolar lavage fluid in different group rats.Results After treatment,compared with those of COPD group,the COPD indexes of the rats in acupuncture group were improved in varying degrees including pulmonary dynamic compliance and expiratory resistance(P<0.01) and pathological inflammation.The IFN-γand IL-4 levels in periphery were significantly decreased after acupuncture treatment(P<0.05,P<0.001).Conclusions Acupuncture treatment has good efficacy on COPD rats and its mechanism might regulate the proportion of Th1/Th2.
