摘  要:目的:观察天麻素对豚鼠噪声性声损伤预防作用及对耳蜗毛细胞SDH影响,同时探讨其可能的作用机理。方法:将实验用雄性红目纯白豚鼠30只,随机编3组,正常组、预防实验组(I_预)、预防对照组(I_对)。将除正常组外2组动物于隔音室内暴露于105dB SPL稳态白噪声下,暴露时间持续每天4小时,连续7天。预防实验组于噪声前3天开始给天麻素100mg/kg,腹腔注射,持续至噪声结束当天,预防对照组同期给予注射用水;测ABR以观察听阈值变化;光镜观察毛细胞琥珀酸脱氢酶(SDH)活性变化。结果:预防实验组ABR听阈值明显低于预防对照组(P<0.05);SDH染色细胞预防实验组明显优于预防对照组。结论:进一步证实天麻素能增加耳蜗毛细胞SDH活性,保护纹血管内皮细胞系统功能。对噪声性声损伤有一定的预防作用。[Objective]To observe the preventing effects of Gastrodia elata Bl on noise - induced hearing loss(NIHL) in guinea pigs and alteration of SDH of hair cells for exploring its probable mechanisms.[Method]Thirty healthy guinea pigs with normal tympanic membrane and auricle reflex were randomly divided into three groups:normal control group,prevention group with Gastrodia elata Bl(I1) and with normal saline(I2);All animals except normal control group were simultaneously exposed to steady white noise at 105dBSPL for 4 hours per day and lasted for 7 days.Animals in I1 began to inject Gastrodia elata B1 (100mg/kg,celiac injection) three days before noise exposure and lasted for 10 days,whereas those in I2 were given normal saline instead of Gastrodia elata B1.Auditory brainstem response(ABR) was used to assess the variation of the auditory function. The variety of succinate dehydrogenase(SDH) of hair cells was observed under light microscope.[Result]Compared with I2,I1 showed significantly lower ABR threshold(P < 0.01),and more SDH - staining hair cells.[Conclusion]Gastrodia elata B1 has obvious prevention effect on NIHL.The mechanisms of the prevention effect of Gastrodia elata B1 are ameliorating the activities of SDH in the hair cells,protecting the functions of the vascular endothelial cell system in the stria vascularis, as well as improving the microcirculation of the cochlea.
