摘 要:目的探讨香丹注射液不同提取部位对麻醉大鼠血流动力的影响。方法将大鼠随机分为7组:生理盐水组、香丹注射液组、香丹注射液石油醚提取部位组(a)、氯仿提取部位组(b)、醋酸乙酯提取部位组(c)、正丁醇提取部位组(d)、水提取部位组(e),给药剂量为16g生药/kg,静脉注射方式给药。用股动脉插管、右颈总动脉心室插管和连续记录肢体Ⅱ导联心电图的方法观察给药前和给药后5、10和15min大鼠的心率(HR)、收缩压(SBP)、舒张压(DBP)、平均压(MAP)、左心室收缩压(LVSP)、左心室终末舒张压(LVEDP)、左心室内压最大上升速率(+dp/dtmax)、左心室开始收缩至dp/dtmax的间隔时间(t-dp/dtmax)和左心室内压最大下降速率(-dp/dtmax)的变化。结果静脉注射相应的受试物后,石油醚提取部位组(a)和乙酸乙酯提取部位组(c)对麻醉大鼠血流动力学的各项指标无显著性影响(P>0.05);氯仿提取部位组(b)能使大鼠SBP、DBP、MAP显著升高(P<0.05);水提取部位组(e)能使大鼠SBP显著升高(P<0.05);正丁醇提取部位组(d)能使大鼠SBP、DBP、MAP、LVSP和-dp/dtmax显著升高(P<0.05);香丹注射液五个不同提取部位对HR、LVEDP、+dp/dtmax和t-dp/dtmax指标无显著性影响(P>0.05)。结论香丹注射液的正丁醇部位提取物具有增强心肌收缩功能和改善心肌舒张功能。Objective To investigate the hemodynamics effect of various extracts in Xiangdan Injection on anesthetized rats.Methods All the rats were randomly divided into seven groups as follows:the saline group,the Xiangdan (XD) Injection group,the petroleum ether extracts in XD group a,the chloroform extracts of the XD group b,the acetic ether extracts of the XD group c,the n-butanol extracts in XD group d,and the water extracts in XD group e.The injection dose of crude drug is 16 g/kg (iv).Changes of hemodynamic parameters including heart rate (HR),systolic blood pressure (SBP),diastolic blood pressure (DBP),mean aortic pressure (MAP),left ventricular systolic pressure (LVSP),left ventricular end-diastolic pressure (LVEDP),left ventricular maximum upstroke velocity (+dp/dtmax),left ventricular maximum downstroke velocity (-dp/dtmax) and the interval from that when the left ventricular begins contracting to the dp/dtmax (t-dp/dtmax) were recorded in 5,10,and 15 min before and after injection by cannulating femoral artery,right common carotid artery,and consecutive electrocardiographic Ⅱ.Results After injection into anesthetized rats,there were no significant hemodynamic changes in group a and c (P>0.05);The SBP,DBP,and MAP rose significantly in group b (P<0.05);The SBP grew up significantly in group e (P<0.05);The SBP,DBP,MAP,LVSP,and-dp/dtmax rose significantly in group d (P<0.05);There were no significant changes with HR,LVEDP,+dp/dtmax,and t-dp/dtmax in XD group (P>0.05).Conclusion The results suggest that the n-butanol extracts in XD group can significantly increase the cardiac muscle contraction of anesthetized rats,and therefore improve their diastolic functions.
