摘 要:从海绵中分离真菌并通过抗肿瘤活性筛选获得活性菌株。从海绵样品中分离得到真菌,并以小鼠白血病P388肿瘤细胞为筛选模型,采用SRB法对所得菌株的发酵产物进行抗肿瘤活性筛选评价,结合薄层色谱和高效液相色谱方法进行化学筛选。从海南采集到的27个海绵样品中分得203株真菌,经化学和生物活性组合筛选得到具有细胞毒活性的菌株15株,选定其中1株高细胞毒活性的菌株作为目标菌株,经鉴定该菌株为黑附球菌Epicoccum nigrum。实验结果表明,海绵来源的真菌是寻找生物活性物质的重要资源,它为抗肿瘤活性先导化合物的发现提供了思路。To isolate sponge-associated fungi and obtain antitumor strains.Isolation of sponge-associated fungi was performed,and antitumor activity corresponding to their cytotoxicity against P388 cells was determined by the SRB method.The secondary metabolites of strains were determined by TLC and HPLC.Two hundred and three strains were isolated from 27 sponge samples collected from Hainan Province.Fifteen strains showed cytotoxicity after combinatorial screening.The target strain,which was identified as Epicoccum nigrum,showed high cytotoxicity.The results indicated that sponge-associated fungi could be one of the most important resources in searching for new bioactive compounds,and it also provides ideas for finding new antitumor lead compounds.
