黄芪注射液对大鼠腹膜细胞凋亡与增殖平衡的作用

摘　　要：目的:探讨中药黄芪注射液对腹膜细胞凋亡和增殖细胞核抗原(PCNA)表达的影响。方法:在正常 SD 大鼠上建立腹膜透析模型,用含黄芪注射液(200 mg/d,400 mg/d)的生理盐水灌注腹腔,20 ml/次,每日2次;用生理盐水作对照。4周后杀检,取腹膜用 LSAB 试剂盒和 TUNEL 试剂盒检测腹膜细胞凋亡和PCNA 表达。结果:黄芪能显著诱导腹膜间皮细胞凋亡,可刺激 PCNA 表达增加,PCNA 表达量呈浓度依赖性。结论:黄芪注射液加入腹透液中可调节腹膜细胞消长平衡。To study the effect of Astragalus Injection(AI)on peritoneal cells apoptosis and proliferative cells nuclear antigen(PC- NA)expression.Methods:The animal model of peritoneal dialysis was established in normal SD rats.AI (20 ml)in different concentra- tions was perfused to the modal,twice a day with daily dose of 200 mg or 400 mg.By the end of the 4th week,the peritoneum was tak- en out,the levels of peritoneal cell apoptosis and PCNA expression were detected immunohistochemically with LSAB kit and TUNEL kit,and compared with those in the control treated with normal saline.Results:Al could significantly induce the apoptosis of peritoneal mesothelial cells and increase the PCNA expression concentration-dependently.Conclusion:AI could regulate the balance between the peritoneal cells apoptosis and proliferation of peritoneal mesothelial cells when added in the peritoneal dialysate.
