摘 要:超排处理 56只昆明小白鼠 ,共得到 488枚胚胎。在室温 ( 2 5℃ )下 ,采用 4种不同的冷冻模式 (常规冷冻、直接冷冻、玻璃化一步法冷冻和二步法冷冻 )进行比较研究 ,并用台盼蓝染色法检测冷冻胚胎质量。结果表明 ,常规冷冻方法最佳 ,平均存活率为 74.88% ,显著优于其余 3种方法 ;玻璃化一步法和二步法差异不显著 ,其平均存活率分别为 6 4.58%和 6 7.70 % ,但显著高于直接冷冻法 ( 50 .50 % )KM mice were randomly divided into three groups and treated for superovulation with PMSG+HCG. 488 mouse embryos were obtained. At the room temperature, embryos were cryopreserved employing four freezing protocols: slow cooling, quick cooling, one-step and two-step vitrification. After thawed in water bath at 37℃, embryos recoved were washed with PBSS solution and stained by Trypan blue to test the quality. The results indicated that the average survival rate of slow cooling (74.88%) was significantly higher than those of other three protocols. The survival rates of one-step and two-step vitrification were 64.58% and 67.70% respectively, and these two protocols were no significant difference, but significantly higher than quick cooling, whose survival rate is only 50.50%.
