高效液相色谱法测定异麦芽低聚糖的组分

摘　　要：采用高效液相色谱法(HPLC),以Zorbax C_(18)柱来分离柱,水为流动相,示差折光仪为检测器,一次进样可同时测定葡萄糖、异麦芽糖、异麦芽三糖和潘糖、异麦芽四糖等。本法对双酶协同作用制取异麦芽低聚糖工艺研究中的低聚糖进行测定,所得结果良好,变异系数为0.28％-1.66％,线性相关系数为0.9996-1.000,低聚糖的检出限在微克级。In this paper, an analytical method for determination of isomaltooligosaccharide by high performance liquid chromalography (HPLC) is presented. The HPLC condition are Zor- bax C_(18)column (4.6mm i.d.×250mm), water as mobile plase and refractive index detec- tor. Glucous、isomaltose、isomaltotriose and panose、isomaltotetraose etc, can be simultane- ously determined in a single injection. The method has been used to determine the isomal- tooligosaccharides in oligosaccharide productions. The results are satisfactory. The CVs were 0.28%-1.66% and correlation cofficients were 0.9996-1.000. The lowest detectable limit for isomaltooligosaccharide was in microgram level.
