摘 要:用酶组织化学和免疫组织化学双标技术,观察了正常SD大鼠基底前脑内侧隔核(MS)、斜角带垂直支(VDB)和水平支(HDB)中NOS阳性神经元的形态和分布及NOS与胆碱能神经元标志物ChAT、NGF受体(NGF-R)和AChE之间的共存关系。结果发现,MS、VDB和HDB的头端NOS阳性神经元较多、胞体较大、突起多,尾端NOS阳性神经元数目较少、胞体较小、突起少而短。NOS+ChAT双标神经元占NOS阳性神经元总数的90%,占ChAT阳性神经元总数的39%;NOS+NGF-R双标神经元占NOS阳性神经元总数的83%,占NGF-R阳性神经元总数的40%;NOS+AChE双标神经元占NOS阳性神经元总数的96%,占AChE阳性神经元总数的39%。这些结果为研究Alzheimer'sdisease病理过程中基底前脑隔区胆碱能神经元退变与NO的关系提供了形态学依据。Using the double labelling technique of enzyme histochemistry and immunohistochemistry, the morphology and distribution of NOS positive neurons and the colocalization of NOS and ChAT, NOS and NGF R, or NOS and AChE in neurons within the medial septum(MS), vertical and horizontal limb of diagonal band of Broca (VDB, HDB) in normal SD rat basal forebrain were studied. The results suggested that there were more NOS positive neurons with larger perikarya and more processes in the rostral region of MS,VDB and HDB, while fewer NOS positive neurons with smaller perikarya, fewer and shorter processes in the caudal region. The double labelling neurons of NOS and ChAT accounted for 90% of total NOS positive neurons and 39% of total ChAT positive neurons. The double labelling neurons of NOS and NGF R accounted for 83% of total NOS positve neurons and 40% of total NGF R positve neurons. The double labelling neurons of NOS and AChE accounted for 96% of total NOS positive neurons and 39% of total AChE positive neurons. These results provide morphological evidence for studying the relationship between degeneration of cholingergic neurons and nitric oxide within septal region in Alzheimer's disease(AD).
