摘 要:本文采用酶细胞化学方法,在光镜下用积分分级计数法,全自动图像分析仪检测精子LDH-C4活性,两法测定LDH-C4活性显著正相关(r=0.801和0.742,P<0.01);LDH-C4酶细胞化学电镜技术观察精子LDH-C4活性,通过计数酶点可测定LDH-C4活性。本文检查38例不育男性和20例已生育男性精子LDH-C4活性,LDH-C4主要分布于精子STM及胞质,其次是顶体及质膜表面。可通过胞膜外逸到精浆,LDH-C4在各部活性比值为5∶2∶1∶1。不育组精子LDH-C4可能通过膜破损处外漏到精浆使精子内LDH-C4显著减少,LDH-C4在精子顶体亦显著减少,使不育组精子LDH-C4活性显著低于生育组精子,提示LDH-C4活性的定位和大小直接与生育能力相关,检测LDH-C4活性可作为检查不育症精子质量的可靠指标。LDH-C4与精子密度显著相关(r=0.737和-0.493,P<0.05);与活动性无相关性。By means of enzymic cytochemistry, the activity of Lactate Dehydrogenase (EC 1.1.1.27) isozyme C4 (LDH C 4) in spermatozoa was determined and measured by adding up the reactive scores. At light microscopic level, the result of accounting scores was significantly positive relevant to that of image analysis ( r =0 801 and 0 742, respectively; P <0 01). At ultrastructural level, activity of LDH C 4 ws determined with enzymic ultracytochemistry. 38 infertile men and 20 fertile men were examined with these methods the results showed that LDH C 4 mainly localized in cytosols and matrix of sperm type mitochondria(STM), partly on acrosome, plasma membrane and some escaped out of membrane. The activity ratio between four areas was 5∶2∶1∶1. The LDH C 4 activity of infertile group was remarkably lower than that of fertile group ( P <0 001). It was observed that the plasma membrane of infertile spermatozoon was damaged and LDH C 4 might flow outside, thus the activity of LDH C 4 in many parts such as acrosome was greatly decreased, and the generation of LDH C 4 in spermatozoon could be relevantly lower than that in fertility. It is proved that the examination of LDH C 4 can serve as relialde item to determine spermatozoon quality. The activity of sperm LDH C 4 is clearly corelated with sperm concentration ( r =0 737 and -0 493, respectively P <0 05) but it has no corelation with sperm motility.
