Inhaled Nitric Oxide at 44ppm Level Reverses Hypoxia-induced Pulmonary Vasoconstriction in the Newbron Rabbit Model

摘  要:目的为吸入一氧化氮治疗新生儿肺动脉高压提供实验依据及经验。方法 30只7~10日龄新生兔随机分3组,观察吸入44 ppm 浓度一氧化氮对急性低氧和正常新生兔肺动咏压的影响。结果吸入5%低氧气3分钟可使新生兔平均肺动脉压升高36.1±15.9%(x±s),下同)。此时吸入44ppm 一氧化氮,经过3、5、7分钟,分别使平均肺动脉压下降20.3±12.3%、25.5±11.4%、28.4±12.5%(与对照组比较 P<0.01),而对心率、血气性低氧性肺动脉高压。Objective To provide laboratory data and direct experience of inhaling nitric oxide (NO) to treat persistent pulmonary hypertension of neonate.Methods Examined the effect of No on pulmonary arterial pressure of inhaled NO while rabbits were inhaled 44ppm NO,5% O_2 and air respectively in this model and those inspired air.Results Pulmonary hypertension was induced by reducing the fraction of inspired oxygen to 5% for 3 minutes.While 44ppm NO was added,a rapid decrease in pulmonary arterial pressure was observed (P<0.001).After 3,5,7 minute,the mean pulmonary arterial pressure (mPAP) decreased by 20.3±12.3%,25.5±11.4%,28.4±12.5%,re- spectively (P<0.001).Inhaled NO didn't influence the mPAP of normal lung.There was no signifi- cant difference noticed in the cyclic guanosine monophosphate concentration,heart rate,artierial blood gas tensions between before and after inhaling 44ppm NO for 7 minutes.Conclusion Shortly inhaled nitric oxide at 44ppm level reverses hypoxia-induced pulmonary vasoconstriction in the new- born rabbit model.
