摘 要:用3—4月龄的治疗性引产胎儿的肝、胸腺制成二种组织浸液,分别加入无关个体混合淋巴细胞培养(MLC)体系中,培养120小时后掺入~3H-TdR,测CPM值,计算刺激指数(SI);用经PCR生物素掺入的HLA-Ⅰ、Ⅱ类特异性探针进行细胞原位杂交,图像扫描定量。与不含胚胎组织浸液的对照组相比,胎肝、胸腺浸液组MLC的SI明显降低,混合淋巴细胞培养的细胞HLA-Ⅰ、Ⅱ类基因mRNA表达均有显著降低,胎肝浸液作用更明显。The supernatant of tissue lysate from liver and thymus of 3-4 month fetus undergoing therapeutic abortion were added to the mixed lymphocyte culturc (MLC) system of unrelated individuals. Compared to the control group, the supernatants of hepatic and thymic lysate could significantly reduce the stimulation index of MLC and HLA-class I and II mRNA expression in the culture celis with cytohybridization in situ technique, employing biotin-labelled cDNA probes. The results suggested that the fetal liver and thymus tissues lysate may inhibit the stimulation between two individuals lymphocytes through reducing cellular HLA genes expres-sion, so that it probably increase the cellular immunotolerance.
