摘 要:重组人粒巨噬系集落刺激因子(rhGM-CSF)连续5天注射对射线及环磷酰胺所致小鼠造血功能低下有明显的改善作用,其疗效与病因、用药途径、用药时机和剂量有密切关系。GM-CSF对7Gy照射小鼠的疗效优于对环磷酰胺致伤小鼠的疗效;腹腔注射的疗效优于皮下注射的疗效;先于环磷酰胺给GM-CSF的效果略胜于同时起用;剂量过大有相反作用,故而提出必须重视GM-CSF的合理使用。文中讨论了GM-CSF通过体内包括造血微环境在内的复杂造血调节网络,主要在造血祖细胞水平影响其增殖分化的机理。The therapeutic effect of GM-CSF on myelosuppression in mice was observed. The myelosuppression was induced by whole-body irradiation and cyclophosphamide treatment. In comparison with solvent controls, recombinant human GM-CSF showed a favorable influence on the recovery of marrow hypoplasia. It was found that the effects of GM-CSF were obvious-ly dependent on the reasons of hypoplasia and on the route of administration, administration schedules and dose of this cytokine. The better effects were obtained in γ-irradiated mice. The more significant promoting effect was found when GM-CSF was given intraperitoneally than subcutaneously. GM-CSF given before the administration of cyclophosphamide generally pro-duced better effects than that given later. Overload of GM-CSF might bring about a negative result in granulopoiesis. According to these experimental results, it seemed reasonable to suggest that a great attention must be paid to the research of optimal application of GM-CSF, especial-ly in clinical use. The mechanism of the effects of GM-CSF might be related to the complicative regulatory network of hematopoiesis including the components of hematopoietic microenvironment. GM-CSF might mainly influence the proliferation and differentiation of hematopoietic progenitor celis and / or the differentiation of hematopoietic steni celis.
