摘  要:10份去血小板的脐血血浆混合后用于研究其对造血细胞培养的支持作用。发现按10%的终浓度加入粒-巨噬细胞集落(CFU-GM)琼脂培养体系时,集落产率稍高于用GM-CSF作刺激因子组。当按30%终浓度加入含EPO的甲基纤维素培养体系时,可支持CFU-GM、爆式红系集落(BFU-E)、巨核细胞集落(CFU-Meg),粗-巨噬细胞与红系细胞混合的集落(CFU-GEM),粒-巨噬细胞与红细胞、巨核细胞混合的集落(CFU-GEMM)生长,而混合成人血浆则无此作用。植物血凝素刺激的单个核细胞条件液(PHA-MNCCM)对大部分集落有促进作用,但均无统计学意义。国产IL-3可提高CFU-GEM产率,但似乎不利于含巨核细胞的集落生长。研究结果提示,脐血血浆含有丰富的造血刺激活性,利用脐血血浆进行造血祖细胞体外集落培养可能为简化并推广体外造血细胞培养提供新的途径。Cord blood plasma from ten cord blood samples were mixed added studied for its role in the colony culture of hematopoietic progenitor celis. While added into the agar culture medium for day-10 CFU-GM at the concentration of 10% in stead of 40 ng / ml of Leucomax (rhGM-CSF), it was able to stimulate CFU-GM growth as well as Leucomax. In methylcellulose culture medium, with the presence of Epoectin-α, 30% of mixed cord blood plasma was found capable of stimulating the growth of kinds of colonies formed by hematopoietic progenitors, including CFU-GM, BFU-E, CFU-Meg, CFU-GEM and CFU-GEMM. In contrast, mixed blood plasma from 4 normai blood donor lacks this ability. PHA-MNCCM could promote the growth of some colonies to some extent, but without significant benefit. IL-3 was able to promote the growth of CFU-GM and CFU-GEM but was found interfering the growth of megakaryocyte progenitors. This result suggests that cord blood plasma may contain rich hematopoietic stimulating activity and that it may help to establish a simple and effective method for in vitro culture of hematopoietic progenitor cells.
