摘 要:目的 探讨硫芥中毒后淋巴细胞谷胱甘肽含量变化。方法 用密度梯度离心法分离大鼠脾淋巴细胞 ,与硫芥共同进行体外培养。用MTT法测定细胞活力。以荧光分光光度计法测定细胞还原型谷胱甘肽 (glutathione ,GSH)、氧化型谷胱甘肽 (oxidizedglutathione ,GSSG)含量变化 ,结果 淋巴细胞活力在硫芥中毒后降低水平与细胞内GSH含量呈直线相关关系。硫芥中毒 8hGSH含量开始下降 ,8~ 12h维持在低水平 ,2 4~ 48hGSH进一步下降 ;GSSG含量 2 4~ 48h显著低于对照水平 ;GSH GSSG比值在 8h后显著降低。Objective To investigate the effects of sulfur mustard on reduced glutathione (GSH) content in rat lymphocytes in vitro . Methods Rat spleen lymphocytes were isolated by density gradient centrifugation and co cultured with sulfur mustard in vitro . The cell viability was detected by MTT method and the contents of GSH and oxidized glutathione (GSSG) in lymphocytes were determined using a fluorometric assay. Results There was a linear correlation between the viability of lymphocytes and the level of GSH after sulfur mustard intoxication. The level of GSH in rat lymphocytes treated with sulfur mustard decreased significantly at 8 h, and this lower level of GSH was maintained until 12 h. A further decline was observed at 24 h. The level of GSSG decreased significantly at 24 h after sulfur mustard treatment. The ratio of GSH/GSSG markedly decreased at 8 h. Conclusion There exits a signi ficant correlation between the viability of lymphocyte and the content of GSH after sulfur mustard intoxication.
