摘 要:TC-1基质细胞系是从小鼠骨髓细胞长期液体培养中分离出来的细胞系,其条件培养液中具有多种生物活性物质,被认为多因子分泌的细胞系。TC-1条件培养液与正常小鼠骨髓细胞共同孵育,然后通过粒、单系祖细胞(CFU-GM)半固体培养试验,脾结节形成试验,放射保护试验及流式细胞仪测定骨髓细胞的周期特点,证实了TC-1细胞系能分泌粒、单系集落刺激活性物质(GM-CSA)及体外对多能造血干细胞具有保存作用。A murine marrow stromal cell line, termed TC-1, secretes various hematopoietic growth factors. Incubation of fresh murine bone marrow cells with TC-1 conditioned medium (CM) in liquid culture resulted in a marked stimulation of hematopoietic cell proliferation. After 24h of culture, there were increased total cell numbers and blast cells in the cell suspension with TC-1 CM compared with the control group. CFU-GM content increased by 18% by 24h of culture with TC-1 CM. In contrast, in the absence of TC-1 CM only 40% CFU-GM remained. The number of day 12 CFU-S (CFU-S12) also increased in the group with TC-1 CM for 24h (No cells: CFU-S12<50; Fresh bone marrow cells: CFU-S12 13.30+1.20/5 × 104MNC; Bone marrow cells treated with TC-1CM: CFU-S12 14.30+1.74 / 5 ×14MNC; Bone marrow cells incubated with culture medium, without TC-1 CM: CFU-S126.10+0.68). The radioprotection assay results showed that the 30 days survival rate was 60-70% when the lethally irradiated mice were injected with 1 ×106 fresh bone marrow cells or bone marrow cells incubated with TC-1 CM for 24h. In comparison, only 20% of the mice survived in the group injected with cells incubated in culture medium without TC-1 CM. Cell cycle analysis of bone marrow cells was performed by flow cytometry. The percentage of S-phase and G2M-phase cells in bone marrow cells incubated with TC-1 CM increased. Taken together, these data indicated that TC-1 cell line may also secrete factors effecting hematopoietic stem cell survivalin addition to CSF.
