Mechanism of the Hypocholesterolemic Effect of CsB-851 (a Fraction Separated from Fermented Mycelium of CordycepS Sinensis) in Mice

摘  要:人工发酵虫草菌丝体有效部分CsB-851能使正常及高脂小鼠血清总胆固醇分别降低10~20%及25~47%。以〔2-(14)C〕-乙酸钠为前体进行参入实验,发现CsB-851使参入小鼠肝胆固醇(游离和酯化胆固醇)的放射性明显减少,但不影响对外源性胆固醇的吸收与排出。提示其降低血胆固醇的主要机制在于抑制肝胆固醇的合成。Cordyceps sinensis (Cs) is a famous Chinese traditional medicine. The ethanol ext-fact of fermented mycelium, CsB, has been reported to be hypolipidemic.Our experim-ents showed that CsB-851, a fraction, separated from CsB, was 5-10 times more active in lowering blood cholesterol than CsB both in normal and in tritpn-treated hyperlipid- emic mice. The serum total cholesterol level was shown to be reduced 10-20% in normal mice and 25-47% in triton Indued hyperlipidemie mice after sc injection of 100-400mg 80/kg. Incubation of sodium acetate-2-14C with liver S10 preparaton from triton-treated mice, showed that the incorperation of radioactivity into liver cholesterol was significa-utly reduced (approximately 50% ) by the treatment with CsB-851. No change in chole-sterol absorption and little increase in: fecal neutral steroids excretion in normal mice were observed after subeutaneous injection of CsB-351 200mg/kg. This suggests that the probable meahanism of the hypocholesterolemic action of CsB-851 is the inhibition of eholesterol biosynthesis,in liver.
