摘  要:目的:观察兔骨髓基质细胞(bonemarrowstromalcells,MSCs)的部分生物学特性及植入心肌后的存活状况。方法:密度梯度离心与贴壁粘附得到高纯度的兔下肢骨MSCs,建立体外培养体系,光镜、电镜、细胞周期等观察其生物学特性,4',6-二脒基-2-苯吲哚(4',6-diamidino-2-phenylindone,DAPI)标记培养的MSCs,植入心肌后检测植入细胞在6周内不同时相点的存活状况。结果:体外培养的MSCs显示良好的贴壁扩增生长能力,88%的培养细胞为静止期细胞,96.4%细胞为活细胞,电镜证明培养细胞显示幼稚细胞的结构。标记的MSCs植入心肌后至少生存6周并且植入细胞扩散融合于植入区宿主心肌中。结论:体外培养的MSCs显示幼稚细胞形态,标记的培养MSCs植入心肌后可以长期存活并扩散融合于宿主心肌。AIM:To observe the biological characters of rabbit bone marrow stromal cells(BMSCs)and its survival status in myocardium. METHODS:The high purity BMSCs were obtained by density gradient centrifugation adhering to the wall of culture flask,and the culture system was established in vitro.The biological characters were observed under light microscopy and electron microscopy.BMSCs were labeled with 4'6 diamidino 2 phenylindole(DAPI),and then implanted into the myocardium.Survival status of implantation cells was observed in 6 weeks at different time point. RESULTS:The BMSCs in vitro demonstrated good amplification ability adhering to wall,88%of which were cells in quiescent period,and 96.4%were living cells.The cultured MSCs observed by electron microscopy displayed immature structure.Labeled MSCs could survive at least 6 weeks after implanting into myocardium and would amplify and amalgamate in the host myocardium. CONCLUSION:The cultured BMSCs in vitro demonstrate immature structure,labeled MSCs can survive for a long time after implantation into myocardium,and diffusedly amalgamate into the host myocardium.
