新疆辣椒上一种球状病毒的鉴定

摘  要:1990年,从辣椒上分离到一个病毒分离物,代号P—9003。人工接种可侵染6科14种植物,回接辣椒产生轻微花叶,不被蚜虫传播,可通过汁液摩擦传染。致死温度80—85℃,稀释限点为10^(-7),体外保活期18天左右。病毒粒体球状,密集时呈多角形,直径33—34nm。该病毒在寄主体内浓度较大,通过差速离心和蔗糖梯度离心容易获得较高浓度的纯化病毒,病毒衣壳蛋白分子量为一个组份,约为27500道尔顿。核酸分子量为三组份,分子量分别为1.24,0.55和0.37×10~6道尔顿。与黄瓜花叶病毒(CMV),蚕豆萎蔫病毒(BBWV),烟草坏死病毒(TNV),烟草环斑病毒(TRSV),香石竹环斑病毒(CRSV)的抗血清不发生反应。与番茄黑环病毒(TBRV)抗血清产生微弱的沉淀线。衣壳蛋白分子由211个氨基酸组成。根据多项测定结果认为,虽P—9003与TBRV抗血清有微弱反应,但其它各项特性与TBRV相差甚远,为非特异反应。P—9003的核酸和蛋白分子量与已知的几个球状病毒组的数据均不相符,因此认为P9003为一种木报导过的球状病毒,暂定名为辣椒轻花叶病毒(PepperMild Mosaic Virus,PMMV)。A virus isolate-P9003 was isolated from pepper in 1990. Infection was obtained by sap inoculated of 14 species in six families. The pepper inoculated by the virus shows a mild mosaic. It can not be transmited by Aphis (Myzus persicae). The virus is infective in Chenopodium amaraticolor sap after dilution in excess of 10-7, after heating for 10 min at 80-85℃ , and on storage for 18 days at room temperature. The virus particle is isometric 33-34 nm in diameter, A dense pure virus preparation was obtained by differential centrifugation and sucrose density gradients. Polyacrylamide gel elec-trophoresis of SDS-treated virus shows one polypeptide of M. Wt. 27500. Three species RNA of M. Wt. 1. 24, 0. 55 and 0. 37×106, estimated by electrophoresis in 1% agarrose gel. The virus was no found serological relationship with cucumber mosaic virus, broad bean wilt virus, tobacco necrotic virus, tomato ringspot virus and carnation ringspot virus in agar gel double diffusion test. The coat protein subunit of the virus is made of 211 molecules of amino acids. According to a lot of studies, we think the virus was not reported. So we named the virus pepper mild mosaic virus (PMMV).
