摘 要:本文采用聚丙烯酰胺凝胶电泳和光密度扫描法测定海拔7m,2300m,4300m健康人红细胞乳酸脱氢酶同工酶活性,酶谱的分离。结果与急性缺氧,低氧适应条件下人血清乳酸脱氢酶同工酶酶谱比较,红细胞呈现了特异酶谱。在扫描积分中,苏州组与西宁组的亚基百分活性及总酶活性相似,而苏州西宁两组与天峻木里组比较亚基相对百分活性及总酶活性有明显差异(P<0.05)。研究结果提示,海拔3000m以下人红细胞乳酸脱氢酶同工酶改变不大,随着海拔的增高,同工酶M型亚基活性增高,酶系统发生相应的代谢调节变化。乳酸脱氢酶同工酶自身调节可能对红细胞的高原缺氧适应起了较为重要作用。This paper presented the assay methods for dissociating erythrocyte lactic dehydrogenase isoenzyme(ELDH-isoenz)activity and zymogram in healthy subjects by polyacrylamide gel electrophoresis and optical density scanning at 7m,2300m and 4300m sea levels. The results were compared with the zymogram of human serum LDH-isoenz under the conditions of acute anoxic and hypoxic adaptation, in which the erythrocytes appeared specificity. In scanning integrals,subunit centi activities and total enzyme activities were showed similarly on Suzhou and Xinging groups, however, both existed significant difference(P<0.05) while they compared with Tianjun-Muli group. It is suggested that the changes of ELDH-isoenz was not defined under 3000m sea level,but following the increasing of sea level,the M type subunit activity of ELHD-isoenz was increased and enzyme system occured corresponding changes of metabolic regulation. The autoregulation of LDH-isoenz might be important effect for anoxic adaptation of erythrocytes at high altitude.
