摘 要:本文首次报道了破伤风毒素及其片段和片段组合对小鼠的免疫效果。通过凝胶过滤和离子交换等方法提纯了破伤风毒素及其片段。各片段中残余素素的污染率为0~0.8%。将各片段给小鼠免疫均能诱导产生保护性抗体,用破伤风毒素攻击后能使部分小鼠免于死亡。当免疫小鼠的血清抗体滴度>1∶4(0.01HAU/ml)时,小鼠在毒素攻击后一般可免于死亡,当血清抗体滴度>1∶128(0.5HAU/ml)时,则小鼠经攻毒后一般不表现出任何中毒症状,当血清抗体滴度处于二者之间时,小鼠一般表现有破伤风中毒症状,但大部分可以存活。毒素的三个片段均能诱导产生保护性抗体,但存在着明显的差别。The protective effect of tetanus toxin,its fragments and the mixture of complementary fragmentswere tested in mice.Tetanus toxin and its fragments were isolated and purified by gel permeasionor ion exchange chromatography.The contamination of residual toxin in each fragment was about 0-0.8%.The immunization with tetanus toxin and its fragments elicited protective antibody responses in mice andprotected the mice from challenge of tetanus test toxin.When serum antibody titer of immunized mice washigher than 0.01HAU/ml,the mice could survive after challenge of test toxin,When serum antibody titerwas higher than 0.5HAU/ml,the mice appeared to be without any signs of tetanus intoxication after cha-llenge.When serum antibody titer was between 0.01 and 0.5HAU/ml,the mice were survived but with thesymptoms of tetanus.It was shown that all of the three fragments of toxin could elicit protective antibodyresponses but with evident difference.
