氮氧自由基化合物对L7712细胞DNA RNA和蛋白质合成的抑制作用

摘  要:2,2,6,6—四甲基哌啶氮氧自由基等自由基化合物,均比其还原体化合物对癌细胞DNA合成的抑制率高。四甲基哌啶氮氧自由基和四甲基吡咯啉氮氧自由基对L7712细胞DNA,RNA和蛋白质合成的抑制作用,在3—24小时内均随作用时间延长而增高,它们对癌细胞DNA合成的抑制作用机理,似属干扰代谢型。Inhibition of 2. 2, 6, 6 - tetramethul - isothiocyano piperdine- 1 -oxyl et al nitroxy radical compounds on the synthesis of DNA in the leukemia 7712 cells in vitro was higher than that of its reduced compounds. The incorporations of [3H] thymidine, [3H] uridine and [3H] leucine into DNA, RNA, and protein respectively in leukemia 7712 cells were inhibited by the comprex of 2. 2, 6, 6 - tetramethy 1 - 4 - isothiocyano-piperidine-1- oxyl and 2, 2, 5, 5 - tetramethy 1 - 4-isocyano-pyrrolion - ocyl with praseodymium dicitrite 50 μg/ml for 3 - 24 hours. The degree of inhibition increased in proportion to the incubation time. After removing the complex from the medium entirely, the rate of DNA synthesis increased with time over 1-7 hours. This result indicated that the inhibition mechanism was likely metabolic lock agent for DNA synthesis.
