灯盏细辛注射液对大鼠肾脏缺血-再灌注损伤的保护作用研究

摘　　要：目的探讨灯盏细辛注射液对缺血—再灌注肾脏损伤的保护作用,为临床防治肾缺血-再灌注损伤提供理论依据。方法 36只SPF级健康雄性SD大鼠,随机均分为3组(n=12)。A组:缺血-再灌注组;B组:灯盏细辛注射液预处理组;C组:假手术组。A、C组大鼠腹腔注射生理盐水,B组大鼠腹腔注射灯盏细辛注射液12 mg/kg.14天后比较3组大鼠肾功能指标、肾抗氧化指标、肾组织细胞凋亡指数以及肾组织Bcl-2、Bax表达情况。结果B组血尿素氮(blood urea nitrogen,BUN)和血肌酐(serum creatinine,SCr)显著低于A组(P<0.05),且显著高于C组(P<0.05);同时,B组超氧岐化酶(superoxide dismutase,SOD)显著高于A组(P<0.05)而显著低于C组(P<0.05),B组丙二醛(malondialdehyde,MDA)显著低于A组(P<0.05)而显著高于C组(P<0.05),肾组织细胞凋亡指数结果显示:B组细胞凋亡指数AI极显著低于A组(P<0.01)而显著高于C组(P<0.01),肾组织Bcl-2、Bax免疫组化染色结果显示:B组Bcl-2表达显著高于A组与C组(P<0.01),B组Bax表达显著低于A组(P<0.05)而显著高于C组(P<0.01),B组Bcl-2/Bax值显著高于A组与C组(P<0.05)。结论灯盏细辛注射液可能通过抗氧化和抗凋亡作用对肾脏缺血-再灌注损伤起到保护作用。Objective To investigate the protective effect of breviscapine on ischemia-reperfusion renal injury, which provides scientific theoretical basis for clinical prevention and treatment of renal ischemia-reperfusion injury. Method 36 SPF male healthy SD rats were randomly divided into 3 groups, 12 rats in each group. Group A was ischemia-reperfusion group, group B was erigeron breviscapus injection preconditioning group, and group C was sham operation group. Rats in group A and C were injected with normal saline, while rats in group B were given 12 ml/kg erigeron breviscapus injection by intraperitoneal injection., After 14 days, renal function, renal antioxidant indexes, renal cell apoptosis indexes and expression of Bcl-2, Bax in renal tissue of three groups were compared. Results The indexes of renal function showed that blood urea nitrogen (BUN) and Serum creatinine (SCr) in group B were significantly lower than group A (P<0.05), but significantly higher than group C (P<0.05). Renal antioxidant indexes showed that superoxide dismutase (SOD) and malondialdehyde (MDA) in group B were significantly higher than group A (P<0.05), but significantly lower than group C(P<0.05). The index of renal cell apoptosis showed that the apoptosis index AI in group B was significantly lower than group A (P<0.01), but significantly higher than group C (P<0.01). The results of immunohistochemical staining showed that the expression of Bcl-2 in group B were significantly higher than that of group A and group C (P<0.05), and the expression of Bax in group B was significantly lower than that in group A (P<0.05) while significantly higher than group C (P<0.01). The ratio of Bcl-2/Bax in group B was significantly higher than group A and group C (P<0.05). Conclusion Erigeron Breviscapus Injection may have a protective role on renal ischemia reperfusion injury though antioxidant and anti-apoptosis.
