摘 要:目的:探讨预先鞘内注射曲马多对福尔马林炎性疼痛大鼠脊髓c—fosmRNA及蛋白质表达的影响。方法:选取鞘内置管成功的32只雄性大鼠随机分成四组,每组8只,分别注射生理盐水(NS组),12.5ug/h曲马多(T1组),25ug/h曲马多(T2组),50ug/h曲马多(T3组)。制备福尔马林炎性疼痛大鼠模型,取模型大鼠脊髓L1~L6阶段脊髓做c—fosmRNA的半定量RT—PCR和Westernblotting检测,观察大鼠脊髓c—fos的mRNA及蛋白质表达量的变化。结果:INS组中的c—fosmRNA表达水平1.34-±0.40明显高于T2组0.69±0.40(P【0.01),高于T1组1.12±0.50,但比较差异无统计意义(P】0.O5);T3组0.41±0.13显著低于T2组,T2组显著低于T1组,其差异均有统计学意义(P【0.01)。Ns组中的c—f0s蛋白表达水平1.20±0.50明显高于T2组0.64±0.40(P【0.01),高于T1组0.98±0.38,但比较差异无统计意义(P】0.05);T3组0.27±0.09显著低于T2组,T2组显著低于T1组,其差异均有统计学意义(P【0.01)。结论:鞘内注射曲马多均能抑制大鼠脊髓背角c-fos的mRNA和蛋白质的表达量;不同剂量的曲马多对cfos表达量的影响不同,但剂量低时对c-fos表达无影响。Objective:To explore the effect of intrathecal pre-injection of tramadol for formalin inflammatory pain in rat spinal cord on c-fos mRNA and protein expression.Methods:A total of 32 male rats with successful intrathecal injection were randomly divided into four groups with 8 in each.All rats were injected with normal saline(Group NS),tramadol 12.5ug/h(Group T1),tramadol 25ug/h(Group T2)and tramadol 50ug/h(GroupT3),respectively.Rat model of formalin inflammatory pain was established.Spinal cord L4-L6 was taken to detect the expression of c-fos mRNA by using semi-quantitative RT-PCR and western-blotting.The changes of c-fos mRNA and protein expression in'rat spinal cord were observed.Results:The expression level of cfos mRNA in Group NS was significantly higher than that in Group T2(1.34+0.40 vs.0.69 ± 0.40,P<0.ODand Group Tl(1.34±0.40 vs.1.12± 0.50,P>0.05) without statistical significance.The expression of c-fos mRNA in Group T3 was significantly lower than that in Group T2(0.41±0.13 vs.0.69 4-0.40),and that in Group T2 was significantlv lower than that in Group T10.69±0.40 vs.1.12±0.50),and there were significant differences(P<0.01).The expression of C—fos protein in Group NS was significantly higher than that in Group T2(l.20±0.50 vs,0.64±0.40,P<0.0l) and Group Tl(l.20±0.50 vs.0.98±0.38.P>0.05) without statistical significance.The expression level of c-fos mRNA in Group T3 was significantly lower than that in Group T2(0.27±0.09 vs.0.64 ± 0.40)and that in Group T2 was significantly lower than that in Group Tl(0.64±0.40 vs.0.98±0.38),and there was significant difference(P<0.01)[Conclusion]Intrathecal injection of tramadol can inhibit c-fos mRNA and protein expression of rat spinal cord dorsal horn.Different dosage of tramadol has different effect on c fos expression,but low dose has no effect.
